Fue construido en el s. II d.C. como mausoleo personal y familiar del emperador Adriano. En el año 403 d.C. quedó integrado dentro de las murallas aurelianas y se convirtió en castillo. Desde el siglo XI se convirtió en propiedad de los papas sirviéndoles de refugio en muchos momentos de la historia. La fortaleza sirvió además como prisión para la Inquisición, alojando a presos ilustres como Giordano Bruno, Galileo Galilei o Alessandro Cagliostro.
